چکیده

2. One method is based on condensation with ethyl acetoacetate in 2N-NaOH to form a product which gives a red colour with Ehrlich's reagent (p-dimethylaminobenzaldehyde). 3. The other method is a modification of the Jaff6 picrate reaction and depends on the reaction of an alkaline picrate complex of ALA with HCI to give an orange-red colour. 4. The effects of varying different factors in the determination, specificity and application to biological materials are described for both methods. I wish to thank Professor A. Neuberger, F.R.S., for his valuable advice and interest in this work, and for the hospitality of his laboratory; Dr J. J. Scott for providing many of the compounds tested in these assays; Mr B. L. Wright, Mr F. Grover andMr 0. Hill for technical assistance.

Urea synthesis by liver homogenates.

The synthesis of citrulline from ornithine and arginine from citrulline has heretofore been associated only with intact liver cells. In the course of studies on the mechanism of the conversion of citrulline to a&nine, a highly active system was found in liver homogenate capable of forming urea in the presence of glutamic acid, adenosine triphosphate (ATP), cytochrome c, an oxygen atmosphere, an...

Transport of ornithine and citrulline across the mitochondrial membrane.

Assay of ornithine transcarbamylase and carbamyl phosphate synthetase I in submitochondrial fractions of rat liver mitochondria shows they are located primarily in the mitochondrial matrix. Therefore during the urea cycle ornithine must pass from the cytoplasm, where it is formed, to the mitochondrial matrix, where it is carbamylated. The resulting citrulline must then pass from the matrix to t...

The effect of various compounds on urea formation by rat liver slices.

In studies on the synthesis of urea in vitro, the nitrogen is usually supplied as NH&l or as (NHJ)$SO~ (l-3). It has been suggested that the mechanism by which such nitrogen enters the cycle and forms urea is somewhat different from that utilized by nitrogen supplied in the form of glutamine or amino acids. Leuthardt and Glasson (2) believe that ammonia from inorganic salts enters the cycle dir...

Metabolic Functions of Biotin Iv. the Role of Carbamyl-l-glutamic Acid in the Synthesis of Citrulline by Normal and Biotin-deficient Rats* by Gladys Feldott

A characteristic consequence of a biotin deficiency in animals is a decreased ability to fix C402 into various tissue components including the amino acid arginine (1). Carbon dioxide is incorporated into arginine as a result of the conversion of ornithine to citrulline in the Krebs-Henseleit urea cycle (2). The methods developed by Cohen and Hayano (3, 4) for the synthesis of citrulline from or...
